MtDNA'nın ND 5/6 gen bölgesi PCR yöntemi ile çoğaltılarak 6 sınırlama enzimi BsurI, AluI, EheI, Hin6I, RsaI, XhoI) ile kesilmiştir. RFLP sonucunda 5 tirsi türünde toplam 13 haplotip saptanmıştır. Türlerin içerisinde ortalama haplotip çeşitliliği 0.6000 ve nükleotit çeşitliliği 0.005331 olarak bulunmuştur. Türlerin kendi aralarında karşılaştırılmalarında ise ortalama nükleotit çeşitliliği 0.008024 ve ortalama nükleotit farklılığı 0.002693 olarak gözlemlenmiştir. Türler arasındaki genetik uzaklık değerlerine bakıldığında en yüksek değerin A. pontica ile A. caspia arasında 0.009599'luk bir değer olduğu, en düşük değerin ise, A. maeotica ve A. tanaica türleri 0.000007 arasında olduğu görülmektedir. Monte-Carlo (X2) İkili Karşılaştırma Analizi sonucu bütün türler arasında önemli derecede farklılıklar olduğu belirlenmiştir (p<0.001). UPGMA soyağacı analizine göre, A. tanaica ve A. maeotica türleri birbirine genetik olarak en yakın bulunmuş, A. pontica ise türler içerisinde en farklı tür olarak göze çarpmıştır</string> <string language="en">In this study mitochondrial DNA polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assay was used to analyse the phylogenetic relationships among five shad species (Alosa caspia, A. fallax nilotica, Alosa maeotica, Alosa pontica, Alosa tanaica) from Turkish seas. The complete NADH 5/6 gene region of mtDNA amplified by PCR were digested with six restriction enzymes, BsurI, AluI, EheI, Hin6I, RsaI, XhoI respectively. A total of 13 haplotypes were detected from five shad species. The average haplotype diversity and nucleotid diversity within species were 0.6000 and 0.005331 respectively. The average nucleotid diversity and nucleotide divergence among species were 0.008024 and 0.002693 respectively. The highest genetic divergence was observed between A. pontica to A. caspia (0.009599) and lowest, between A. maeotica and A. tanaica (0.000007).
