Descriptions : Turkish Bu çalışmada, P. capsici'ye karşı biberlerde (Capsicum annuum L) genetik dayanıklılık konusu araştırılmıştır. Çalışma dört bölüm halinde yürütülmüştür. İlk aşamada yerli ve yabancı 35 farklı biber genotipi, sekizi ülkemizden izole edilen biri ABD'den getirtilen, P. capsici'nin 9 izolatı ile test edilmiş, genotip x izolat interaksiyonu araştırılmıştır. Deneme bulguları, ülkemizde P. capsici'nin fizyolojik ırklarının mevcut olduğunu ortaya koymuştur. İkinci bölümde, P. capsici'ye dayanıklı bulunan dört farklı genotip kendi aralarında melezlenerek bunlardan daha dayanıklı genotipler geliştirilmeye çalışılmıştır. Elde edilen F2 populasyonlarında mevcut ebeveynlerden daha dayanıklı transgresif bireyler elde edilmiş ve bunların kendilenmesiyle de dayanıklı hatlar geliştirilmiştir. Üçüncü bölümde, P. capsici'ye karşı kısmi dayanıklı yerli bir genotip olan KM2-11'deki dayanıklılığın, agresivite düzeyleri farklı üç izolata karşı dayanıklılığın kalıtımı incelenmiş, izolatlara ve hastalığın dönemlerine göre, dayanıklılığın kalıtım modelinin, değiştiği belirlenmiştir. Çalışmanın son aşamasında ise P. capsici'ye karşı dayanıklı ve duyarlı genotiplerden bazılarının filogenetik ilişkileri moleküler tekniklerle (SSR ve SRAP) araştırılmıştır. Bu çalışmada elde edilen bulgular sonucunda genel olarak dayanıklı genotiplerin akrabalık düzeylerinin uzak olduğu; KM2-11 genotipi, genetik olarak yine aynı populasyondan seçilen duyarlı genotip KMAE-12 hattına yakın akraba olduğu belirlenmiştir.
English This study encompasses an investigation of inheritance of resistance against P. capsici Leon on pepper. The study was carried out at four major steps. The first step was to test 35 different pepper genotypes obtained domestically or brought from abroad with nine P. capsici isolates, eight of which originating from Turkey and one from USA in order to study genotype x isolate interactions. Our findings revealed the existence of physiological races of P. capsici in Turkey. The second step was to develop transgressive segregants for enhanced resistance against P. capsici by cross breeding out of a population. The resulting four resistant genotypes identified in the first part of the study were intercrossed and transgressive segregants with enhanced resistance were selected and selfed, giving rise to resistant lines useful for crop improvement. The third step was to investigate the heritability of resistance of the partially resistant domestic genotype KM2-11 which was tested with three isolates with varying virulence levels. It was shown that heritability values changed depending on the isolates and the developmental stages of the disease isolates. The fourth and the last step was to investigate the phylogenetic relationship of pepper genotypes, both resistant and susceptible ones, via molecular techniques (SSR and SRAP). It was shown that resistant genotypes were genetically diverse and that the partially resistant KM2-11 genotype was genetically close to the susceptible "KMAE-12" genotype.
German Diese Studie umfasst eine Untersuchung der Vererbung des Widerstands gegen s. Capsici Leon auf Pfeffer. Die Studie wurde an vier Hauptschritte durchgeführt. Der erste Schritt war 35 verschiedene Pfeffer-Genotypen, die im Inland oder aus dem Ausland mit neun s. Capsici gebracht testen isoliert, acht von denen mit Ursprung aus der Türkei und eine aus den USA um Genotyp x Eingreiftruppe Interaktionen zu untersuchen. Unsere Erkenntnisse offenbart die Existenz der physiologischen Rennen des p Capsici in der Türkei. Der zweite Schritt war, transgressiven Segregants für verbesserte Resistenz gegen s. Capsici durch cross-Zucht von einer Bevölkerung zu entwickeln. Die resultierenden vier resistenten Genotypen identifiziert im ersten Teil der Studie wurden intercrossed und transgressiv Segregants mit verstärkten Widerstand wurden ausgewählt und Selfed, was zu resistente Linien nützlich für die Verbesserung der Ernte. Der dritte Schritt war die Erblichkeit des Widerstands des teilweise resistent inländischen Genotyps KM2-11 zu untersuchen, die mit drei Isolate mit unterschiedlicher Virulenz Ebenen getestet wurde. Es wurde gezeigt, dass Heritabilität Werte je nach Isolate und die Entwicklungsstadien der Krankheit Isolate geändert. Die vierte und letzte Schritt war es, die phylogenetische Beziehung Pfeffer Genotypen, beide resistent und anfällig sind, über molekulare Techniken (SSR und SRAP) untersuchen. Es wurde gezeigt, dass resistente Genotypen genetisch vielfältig waren und die teilweise resistent KM2-11-Genotyp genetisch nahe der Genotyp anfällig "KMAE-12" war.
