Descriptions : Turkish Araştırmada, Manisa lalesi olarak bilinen doğal Anemone coronaria var. coccinea’da anter kültürü yöntemiyle haploid bitki elde edilmesi üzerine çalışılmıştır. Bu amaçla, Adana’da doğal yayılış gösteren bir populasyondan sağlanan eksplantlar, farklı temel besin ortamları ile farklı hormon konsantrasyonları ve ön uygulamalara tabii tutulmuşlardır. Ortam denemelerinde; NN ve B5 temel besin ortamlarına, farklı konsantrasyonlardaki hormonların tek başlarına veya farklı dozlardaki aktif karbon, gümüş nitrat, L-glutamin ile bunların karışımına eklenmesiyle oluşturulan farklı uygulamaların embriyo oluşturma performansları saptanmıştır. Hormon denemelerinde, IAA (0, 0.1, 0.5, 1 ve 2 mg/l) tek başına veya 0.1 mg/l BAP ile birlikte kombinasyon halinde kullanılmıştır. NN ve B5 ortamlarında gerçekleştirilen bu uygulamalar arasında en yüksek embriyo oluşum oranı % 12.7 ile NN ortamındaki 1 mg/l IAA + 0.1 mg/l BAP hormon kombinasyonundan sağlanırken, bunu % 9.3 oran ile B5 ortamındaki 0.5 mg/l IAA + 0.1 mg/l BAP kombinasyonu takip etmiştir. Gümüş nitrat denemeleri (kontrol, 2.5, 5, 10 ve 15 mg/l) içerisinde en yüksek embriyo oluşumu, % 3.3 ile B5 ortamındaki 5 mg/l gümüş nitrat + 0.5 mg/l IAA + 0.1 mg/l BAP içerikli uygulamadan elde edilmiş olup, aynı içerikli NN ortamındaki anterlerden meydana gelen embriyo oluşumu % 2.5 oran ile ikinci sırada yer almıştır. L-Glutamin denemelerindeki (800 mg/l) tek olumlu tepki, oldukça düşük bir oran (% 0.7) ile B5 ortamındaki 800 mg/l L-glutamin + 2 mg/l IAA + 0.1 mg/l BAP uygulamasından elde edilmiştir. Aktif karbon denemelerinin (kontrol, % 0.1, % 0.5 ve % 1) hiçbirisinden olumlu cevap alınamamış, bununla birlikte ortama girdiği aktif karbon + gümüş nitrat + L-glutamin karışım denemelerinde de yine tüm ortamlarda polen embriyogenesisine olumsuz etkide bulunmuştur. Çalışmanın başlangıcında uygulanan soğuk muameleleri (kontrol, + 7 0 C’de 2, 7 ve 21 gün) arasında fark bulunamamıştır. Yapılan sitolojik incelemelerde ise embriyojenik bölünmelerin simetrik, generatif ve vejetatif çekirdek bölünmelerinden kaynaklandığı tespit edilmiştir. Direkt polen embriyogenesisinin amaçlandığı bu araştırmada ayrıca, organogenesis ve oldukça yüksek oranlarda kallus oluşumu meydana gelmiştir.
English Anther culture method was used in Anemone coronaria var. coccinea, known as poppy anemone, in order to obtain haploid embryo production in this study. For this aim, the effect of different media and cold treatments on the explants picked from a red-flowered wild population in Adana were investigated. In culture media experiments, NN and B5 media were used as basic medium. Different hormone concentrations were added to these media either alone or accompanied with activated charcoal, silver nitrate, L-glutamine and their mixture in different concentrations. Then, pollen embryogenesis performances of these treatments were evaluated. In hormone experiments, IAA (0, 0.1, 0.5, 1 and 2 mg/l) was used either alone or together with BAP (0.1 mg/l). Among the treatments in NN and B5 media, the highest pollen embryogenesis with 12.7 % was obtained from 1 mg/l IAA + 0.1 mg/l BAP combination in NN medium. This treatment was followed by 0.5 mg/l IAA + 0.1 mg/l BAP combination in B5 medium with 9.3 %. Among silver nitrate treatments (0, 2.5, 5, 10, 15 mg/l), the highest embryo formation (3.3 %) was acquired from a treatment in B5 medium containing 5 mg/l silver nitrate + 1 mg/l IAA + 0.1 mg/l BAP while the second best treatment (2.5 %) had the same content but in NN medium. In L-glutamine experiments, the single positive reaction was obtained from a treatment in B5 medium containing 800 mg/l L-glutamine + 2 mg/l IAA + 0.1 mg/l BAP with quite low ratio (0.7 %). None of activated charcoal treatments (control, 0.1 %, 0.5 % and 1 %) gave positive result. However, activated charcoal has negatively affected pollen embryogenesis performances in the mixture media in which it was included. It could not be found any difference among the cold treatments (control, 2,7 and 21 days in + 7 0 C) tested in the beginning of the study. In the cytological observations, it was determined that embryogenic divisions had derived from simetric, generative and vegetative nucleus divisions. Additionally, organogenesis and callus formations were realized apart from direct pollen embryogenesis in this study.
German Anthere Kultur-Methode wurde in Anemone coronaria var. coccinea, bekannt als Mohn verwendet. Anemone, um haploide Embryo-Produktion in dieser Studie zu erhalten. Für dieses Ziel, die Wirkung von unterschiedliche Medien und kalt Behandlungen auf die Explantaten nahm rot blühenden wilden Bevölkerung in Adana wurden untersucht. In Kulturmedien Experimenten wurden NN und B5 Medien als basischem Medium verwendet. Verschiedene Hormonkonzentrationen wurden an diese Medien entweder allein oder mit Begleitung aktiviert Kohle, Silbernitrat, L-Glutamin und ihre Mischung in verschiedenen Konzentrationen. Dann, pollen Leistungen der Embryogenese dieser Behandlungen wurden bewertet. In Hormon-Experimenten, IAA (0, 0.1, 0.5, 1 und 2 mg/l) wurde verwendet, entweder allein oder zu zweit mit BAP (0,1 mg/l). Zwischen den Behandlungen in NN und B5-Medien, die höchste Pollen Embryogenese mit 12,7 % wurde von 1 mg/l IAA + 0,1 mg/l BAP Kombination NN Medium erlangt. Dies Behandlung gefolgt von 0,5 mg/l IAA + 0,1 mg/l BAP Kombination an B5 Medium mit 9,3 %. Unter Silbernitrat Behandlungen (0, 2,5, 5, 10, 15 mg/l), die höchste Embryo Bildung (3,3 %) wurde von einer Behandlung mit 5 mg/l Silbernitrat + 1 mg/l IAA + 0,1 mg/l B5-Medium erworben. BAP, während die zweitbeste Behandlung (2,5 %) die gleiche Inhalte, sondern in NN Medium hatte. L-Glutamin-Experimente wurde die einzige positive Reaktion von einer Behandlung in B5 übereignet. Medium mit 800 mg/l L-Glutamin + 2 mg/l IAA + 0,1 mg/l BAP mit sehr niedrigen Verhältnis (0,7 %). Keiner der Aktivkohle-Behandlungen (Steuern, 0,1 %, 0,5 % und 1 %) gaben positives Ergebnis. Jedoch hat Aktivkohle negativ beeinflusst, Pollen Embryogenese Aufführungen in den Mischung Medium, in denen es aufgenommen wurde. Es konnte nicht gefunden werden, keinen Unterschied zwischen kalten Behandlungen (Steuern, 2,7 und 21 Tage in + 7 0 C) am Anfang der Studie getestet. In die zytologischen Beobachtungen wurde festgestellt, dass embryogenic Unternehmensbereiche abgeleitet war von Simetric, generative und vegetative Nucleus Divisionen. Darüber hinaus Organogenese und Hornhaut Formationen bewahrheiteten sich neben direkten Pollen Embryogenese in dieser Studie.
